There's a tiny bit of Gamzee's blood in the panel he disappears in, so I'm assuming he killed himself or something like that.

As far as Dave and Rose's relationship goes as brother and sister honestly if you go through their interaction they're freakin' adorable as siblings. Just that level mix of trust/cajoling. If they weren't related sure, that could work out like that. But they're even better as siblings.
