Yes. Armor penetration is calculated according to all those parameters, angle, etc.
Howitzer's shells will hit, for example, top armor
And it will matter what part of a tank you hit.

Also, how upgradable/customizable are the tanks? For example, if I research better welding (so that they can hold the larger stress of a larger cannon firing) will I be able to change the cannon with a higher calibre one (within reason of course), or will the upgrades focus more on enhancing the performance of what you already have?

By the time of Western release, there will be Tank-killing bombers. Plus LONG-range artillery support.
We are thinking about other NPC additions, but that will happen after the initial release this fall.

In this game, you do not upgrade a single gun. You replace your current gun/hull/turret/chassis/radio with new ones (with better parameters). So, yes, most of vehicles start with basic (smallest cal.) gun and then you replace them with higher caliber (where applicable).
You can also use different types of ammo for each gun, like armor piercing, etc.
Tank crew members - they affect your tank's performance parameters such as firing accuracy, etc.
Plus, the player himself is levelling up and gives some bonuses to certain parameters.
How about anti-tank guns like the Pak 38 or 57mm ZiS 2 to defend "bases"?

Out of what I gather, no you won't be able to modify it that much, but you should be able to mount a larger cannon (Panther had a 75mm, you should be able to mount a 88mm, though probably not much bigger, not a Maus 152mm cannon for example). But I'm not a developer so to be sure wait for their answer.
Well, dual 88mm is a little too much for Panther's chassis. Coelian was supposed to have mere 37 or 55mm duals.

IS-4 and IS-7? I'm guessing it's the Russian response to the Maus, right? But if the IS-3 is the equivalent of the Tiger II (King Tiger), wouldn't the IS-4 be the response to the Maus, and IS-7 something bigger? At least if they are ranked by tier of power (as in size of cannon and armour). Or is the IS-4 somewhere inbetween the Tiger II and the Maus?

Oh, just remembered the Tiger-Maus, is the IS-4 will be the equivalent of the Tiger-Maus I guess (if it's in the game).
IS-3 is more or less equivalent to Tiger II (though IS-3 is a little better in some aspects)
IS-4 is a tough target for Tiger II. Tiger II need a lot of help from his friends (Hummels, JandPanzers, etc) to crack IS-4. It has hard time piercing IS-4's frontal armor.
IS-7 is more of a Maus opponent.

But the most important thing to keep in mind about World of Tanks is this: it's not about "Level 6 tank beats Level 5". Tanks have dozens of parameters, and it's a team battle with huge importance of terrain. So victory goes to the team which co-ordinates well or to more agile and smart tacticians.
Believe it or not, light tanks, like Czhech t38 or Russian BT-2 are the most dangerous enemies of all heavy and super heavy tanks, as they can recon their positions and "light them up" for heavy artillery (Hummels, SU-14, etc.)

THe jagdpanther is probably the highest tier anti-tank for the germans, or else pretty high up, I'm guessing.
As Vittorio said, all tanks are useful, as they all have their role. I haven't played World of Tanks (of course), but in Men of War an M8 Greyhound that gets behind your frontline will do much more damage than a few King Tigers or IS-3s (on average), and since both games aim for realistic (within reason) tanks I guess it will be the same in WoT. I'm guessing most matches will go like this:
Super-heavies: Will define the frontline, they'll be mostly immobile (unless heavily targeted by artillery) shelling each-other at long range from the front without doing too much to each other (they will focus more on not letting the opposing supers turn around and move than actually destroying them).
Heavies: More or less the same as supers, just that they will aim for other heavies and keep the mediums at bay.
Mediums: Will keep mediums and lights away from the bigger ones while keeping away from the frontline (and therefore keeping away from the larger cannons of heavies and supers), possibly following the lights and trying to get behind the enemy on some occasions.
Lights: Will try to get behind the enemy (and once they do...).
Artillery: Will stay put and hidden until they get their sight onto something worth giving away their positions for (heavies, supers and other artillery mostly), then they'll fire away as fast as possible, then full speed to a new position before the other artillery counter-fires.
Organized clans will probably have more advanced strategies, but I believe most battles will go somewhat like the above.
Anyway this is one of the games I'm most hyped for this year.
Above is one of dozens and hundres of possible sets of principles. In World of Tanks teams of 30 vehicles each are pitched against each other on dozens of various maps. Map type and lay out changes everything. Our designers make sure there are no two maps alike (in terms of tactical schemes to be employed). You need to try :-)

M4A5 Sherman existed.
No US vehicle was ever built with the M4A5 designation. It was simply a number to signify Canadian Production.
Since we are playing the US here, they didn't have anything called an M4A5, therefore it doesn't exist.
